Job Title: Sanger Sequencing Specialist

Role Overview:
We are seeking a highly skilled and detail-oriented Sequencing Specialist with hands-on expertise and experience in Sanger sequencing. Experience in Next-Generation Sequencing (NGS) workflows and data analysis is a plus. The ideal candidate will be responsible for performing end-to end Sanger sequencing runs, also preparing NGS libraries, analyzing data, and ensuring accurate and timely delivery of results for internal and client projects.
Key Responsibilities:
- Perform Sanger sequencing workflows, including PCR amplification, purification, and capillary electrophoresis etc.
- Prepare SOPs, engage in Sequencing method development, Operate and maintain sequencing instruments (e.g., ABI 3730xl for Sanger; Illumina or similar platforms for NGS).
- Ability to prepare NGS libraries for various applications (DNA, RNA, targeted panels, amplicon sequencing) for outsourcing.
- Perform quality control checks on libraries and sequencing data using tools like Bioanalyzer, Qubit, and qPCR.
- Analyze sequencing data:
- For Sanger: Interpret chromatograms, base calling, and sequence alignment.
- For NGS: Perform primary and secondary analysis (FASTQ QC, alignment, variant calling).
- Troubleshoot technical issues and optimize protocols for improved efficiency and accuracy.
- Maintain detailed documentation of experiments and results in compliance with laboratory standards.
- Collaborate with cross-functional teams to support molecular biology and genomics projects.
- Ensure adherence to quality control and safety guidelines in the laboratory.
- Participating in various laboratory operations and software development for sequencing requirements.
- End-to-end responsibility of maintaining the sequencing facility but not limited to ordering reagents & spare parts, calibration, PM, repair, training etc.
Qualifications & Skills:
Education: M.Sc./M. Tech in Molecular Biology, Biotechnology, or related field.
- Experience:
- 4-6 years of hands-on experience in Sanger sequencing and similar workflows.
- Experience in NGS library preparation and sequencing platforms (Illumina, Ion Torrent, etc.) is a plus.
- Familiarity with sequencing analysis software and bioinformatics tools (e.g., Chromas, Sequencing Analysis, FASTQC, BWA, GATK etc.).
- Strong understanding of PCR, primer design, and DNA/RNA purification techniques and equipment.
- Excellent troubleshooting and problem-solving skills.
- Ability to work independently and manage multiple projects simultaneously.
- Good communication and documentation skills is a must.
Preferred:
- Experience with high-throughput sequencing and automation platforms.
- Knowledge of advanced bioinformatics pipelines for variant analysis and annotation.
